Peripheral blood mononuclear cells (PBMCs) from a healthy donor were stained with a panel of 31 TotalSeq™-B antibodies (BioLegend).
PBMCs are primary cells with relatively small amounts of RNA (~1pg RNA/cell).
Libraries were prepared following the Chromium Next GEM Single Cell 3ʹ Reagent Kits v3.1 with Feature Barcode technology for Cell Surface Protein User Guide (CG000206 RevA).
- 5,527 cells were detected
- Sequenced on Illumina NovaSeq with approximately 30,853 reads per cell
- 28 bp Read 1 (16 bp Chromium barcode and 12 bp UMI)
- 91 bp Read 2 (transcript)
- 8 bp i7 index (sample barcode)
- run with
--expect-cells=5000
